The aim of this work is to investigate the effect of respiratory chain complex I inhibitors on regulation of mitochondrial permeability transition pore, mitochondrial functions and cell viability in rat brain cortex and cerebellum during global ischemia. Objectives and tasks of the study. 1. To assess the direct effect of complex I inhibitors (rotenone, amobarbital, pentobarbital, metformin and phenformin) and cyclosporine A on isolated cortex and cerebellum mitochondrial functions: Ca²⁺ -induced opening of permeability transition pore and respiration rate by oxidizing complex I and II substrates. 2. To investigate the direct effect of complex I inhibitors (rotenone and amobarbital) and cyclosporine A on Ca²⁺ -induced permeability transition pore opening of cortex mitochondria isolated from rats of different age (5-7 days, 2-3 months, 7-10 months, 24 months). 3.To investigate the effect of intravenous infusion of rotenone and pentobarbital (along with ketamine) on rat brain cortical and cerebellar mitochondrial functions and cell viability after 2 hours of global ischemia.
